Who cares if it spawned a bunch of comedies? The truth is, almost none of the "comedies" are actually funny (besides Napoleon Dynamite, of course).

No, I do not think the humor of Homestar runner is similar to Monty Python.

The general consensus seems to be a resounding "No", but it doesn't look like anybody's broken down the specific rationale, so allow me...

Both have tons of running gags and recurring characters

This could be said about a brazillion other web toons, TV shows, movie series, etc. It doesn't merit any sort of connection between two specific examples from entirely different genres.

Both are absurdical

Sadly, absurd humour that's actually funny is becoming more rare (anyone who says "Family Guy" gets a fork in the eye), but it's still common and varied enough that it also doesn't merit a connection. The vast majority of toons on Weebls-Stuff.com is shockingly absurd, but any Python connections are likely coincidental or require a major stretch (or are blatant tributes).

The Heavy Lourde gag is totally taken from the 16-ton weight from MPFC

That large weight has been a staple of cartoon visual gags since before color TV, and was a visual gag that was well-known before Monty Python even existed, and the Python troupe simply incorporated that device into their works. Obviously it's a matter of opinion, but I think that it's much more likely that its appearance in the animated H*R universe is a nod to the classic cartoons than to any Python gag.
